📌 Indications:

Ruxotor (Ruxolitinib) is indicated for the treatment of:

  • Myelofibrosis (MF): including primary myelofibrosis, post–polycythemia vera MF, and post–essential thrombocythemia MF.

  • Polycythemia Vera (PV): in patients resistant or intolerant to hydroxyurea.

  • Graft-versus-Host Disease (GVHD): in steroid-refractory acute and chronic GVHD.


💊 Pharmacology:

  • Ruxolitinib is a Janus-associated kinase (JAK1 and JAK2) inhibitor.

  • It works by reducing overactive JAK-STAT signaling, which is involved in abnormal blood cell production, inflammation, and immune system dysregulation.

  • By inhibiting JAK1/JAK2, Ruxotor decreases splenomegaly, alleviates disease-related symptoms (fever, night sweats, weight loss), and improves quality of life in MF and PV patients.


💉 Dosage & Administration:

  • Typical starting dose (MF/PV): 5–20 mg orally twice daily, based on platelet count.

  • GVHD: 5–10 mg twice daily (dose adjustments may be needed).

  • Doses should be individualized according to platelet counts, hemoglobin levels, and neutrophil counts.

  • Tablets should be swallowed whole, with or without food.


⚠️ Precautions & Warnings:

  • Monitor for cytopenias (anemia, thrombocytopenia, neutropenia).

  • Risk of serious infections (TB, herpes zoster, hepatitis B reactivation).

  • May increase the risk of skin cancers and non-melanoma malignancies.

  • Regular monitoring of complete blood counts (CBC) is essential.


❌ Contraindications:

  • Hypersensitivity to Ruxolitinib or formulation components.

  • Severe hepatic impairment (without close monitoring and dose adjustment).


🤰 Pregnancy & Lactation:

  • May cause fetal harm; avoid during pregnancy.

  • Breastfeeding is not recommended during treatment.


⚕️ Side Effects:

Common:

  • Anemia, thrombocytopenia, neutropenia

Serious:
